Since dog-ownership practices can affect animal welfare, it is important to understand the level of owners’ awareness of animal needs and current legislation. Therefore, this study aimed to describe dog owners’ knowledge and practices and determine their association with behavior and health problems in dogs. An online survey was conducted, which included questions about responsible ownership practices and the level of awareness of national animal welfare and pet ownership legislation. Descriptive statistics and ordinal multivariate logistic regression analyses were performed. A total of 5,216 individuals responded, of which 3,809 were dog owners. Twenty percent of dogs were described as being overweight or obese, and dogs that were not fed leftovers were less likely to be obese. Additionally, neutered bitches were less likely to be obese than castrated male dogs. In relation to aggressive behavior, 24.6% of owners declared that their dog had shown aggressiveness toward people and 51.1% declared aggressiveness toward other animals. Taking dogs into basic training classes, not taking them out for walks, and living on a farm were all risk factors for aggressive behavior. In terms of owners’ awareness of the current legislation, being moderately or not informed increased the likelihood of not having their dogs identified with a microchip and registered, not keeping their vaccinations updated, or not desexing. An effective educational strategy that uses a multidisciplinary approach is required to improve pet ownership practices in Chile.

- ItemEffect of CRISPR/Cas9 Targets Associated with Iron Metabolism and Its Variation on Transcriptional Regulation of SHK-1 Cell Line as a Model for Iron Metabolism(2024) Dettleff Faundes, Phillip James; Yehwa Jin; Carolina Peñaloza; Rodrigo Pulgar; Sáez Beltrán, Alejandro José; Diego Robledo; Escobar Aguirre, Sebastián GonzaloIn this study, we investigated the function of a gene associated with iron metabolism using CRISPR-Cas9 and RNA sequencing in SHK-1 salmon cells. Our objective was to understand how different guide RNA (gRNA) sequences against the transferrin gene tf could influence gene expression and cellular processes related to iron uptake. RNA-Seq analysis was performed to evaluate the transcriptomic effects of two distinct gRNA targets with high knock-out (KO) efficiencies for the targeted tf gene in the SHK-1 genome. Our results showed no significant differential expression in transferrin-related transcripts between wild-type and CRISPR-edited cells; however, there were major differences between their transcriptomes, indicating complex transcriptional regulation changes. Enrichment analysis highlighted specific processes and molecular functions, including those related to the nucleus, cytoplasm, and protein binding. Notably, different sgRNAs targeting tf might result in different mutations at DNA levels in SHK-1 salmon cells.
- ItemEfficient transfection protocol of rainbow trout gills epithelial (RTgill-W1) cell line through nucleofection system. v1(2024) Escobar Aguirre, Sebastián Gonzalo; Escorza Sius, Amanda Paz; Escobar Aguirre, MatíasIn this protocol, we optimize the experimental condition to express plasmid DNA in salmonid fish rainbow trout gills epithelial (RTgill-W1), using a nucleofection system. The principle of nucleofection is a combination of electrical parameters that ensures efficient DNA delivery into the nucleus, combined with low toxicity and high cell viability. Using the Neon Transfection System (Invitrogen‱ MPK5000; code: 10431915) and the Invitrogen‱ Neon‱ Transfection System Kit 10 μl (brand: Invitrogen‱ MPK1096; code: 10124334) we were able to express over 60% of GFP in RTgill W 1 line cells. These results were achieved after trying different optimization steps voltage (ranges between 850 - 1600 volts), pulse exposure (times 10 30 ms), and number of pulses, being the condition 1600 volts, 20 ms 1 pulse, and 1 μg μL of plasmid the most effective compared to the other conditions, presenting a higher percentage of GFP expression and lower mortality rate.

Facultad de Agronomía e Ingeniería ForestalGran parte de las pérdidas de la industria de salmonicultura son asociadas a la Piscirickettsia salmonis, agente causante de la Septicemia Rickettsial Salmonídea. Para lidiar con este patógeno la industria emplea vacunas y antibióticos, los cuales han demostrado ser insuficientes. Estudios previos han identificado a través de análisis genómico, un grupo de genes candidatos, en familias con resistencia natural al SRS, que podrían estar involucrados en generar esta inmunomodulación. Sin embargo, la mayoría de estos estudios se realizan bajo condiciones controladas. Por lo tanto, este trabajo apuntó a evaluar la respuesta de las vacunas en función de ciertos genes candidatos, bajo condiciones productivas, es decir, balsas jaula en mar. La metodología implicó medir el desempeño de 3 tratamientos con vacunas inyectables, uno de estos sin tener antígeno para SRS como vacuna control, otro con una formulación experimental y el último con la estrategia más empleada en la industria. Se muestrearon 5 animales por grupo desde marzo hasta agosto, extrayendo el riñón cefálico y evaluando el ARN mensajero mediante qPCR con partidores para genes involucrados con metabolismo del hierro y la respuesta inflamatoria. Los resultados indicaron que la expresión de Transferrina y TNF-α aumentan su expresión en el mismo tratamiento que posee menores niveles de mortalidades. Por ende, se demostró que las vacunas tienen un efecto de protección ante el SRS, además se observó una relación entre la expresión de genes relacionados con el metabolismo del hierro y la respuesta inflamatoria.
