Inchin-ko-to (ICKT) is an herbal medicine used in Japan to treat jaundice and liver fibrosis. We investigated the effect of oral ICKT supplementation on endotoxin-induced cholestasis in the rat. Material and methods. Lipopolysaccharide (LPS) injection (1 mg/kg body weight i.p.) was used as a model of sepsis-induced cholestasis. Bite flow, biliary bile salt secretion, biliary glutathione secretion and protein expression of the main hepatobiliary transporters Na(+)-taurocholate-cotransporting peptide (Ntcp), multidrug resistance protein 2 (Mrp2) and bile salt export pump (Bsep) were analyzed by conventional techniques in ICKT treated and non-treated animals. Results. Injection of LIDS induced a significant decrease of bite ftow (-24%), biliary bite salts (-40%) and glutathione excretion (-70%) as well as a significant decrease in Ntcp (-90%) and Mrp2 (-80%) protein levels. ICKT supplementation partially prevented the effects of LIPS determining a less intense reduction in bile flow (-10%), a normalization of glutathione excretion as well as a significant increase in Mrp2 protein levels to 60% of the levels observed in control animals. ICKT administration did not modify the effects of LPS on BS secretion or Ntcp protein levels. Conclusion. Our data show that oral. supplementation of ICKT partially prevents LPS-induced cholestasis by increasing Mrp2 protein levels and biliary glutathione excretion thus increasing bite salt-independent ftow.

The persistence of dictatorship mayors(2021) Gonzalez, Felipe; Munoz, Pablo; Prem, MounuWe look at Chile's transition to democracy in 1990 to study the persistence of authoritarian politics at the local level. Using new data on the universe of mayors appointed by the Pinochet dictatorship (1973-1990) and leveraging on the arbitrary election rules that characterized the first local election in 1992, we present two main findings. First, dictatorship mayors obtained a vote premium that is larger among the last wave of incumbents and appears partially explained by an increase in local spending. Second, dictatorship mayors who were democratically elected in 1992 brought votes for the parties that collaborated with the dictatorship in subsequent elections held in democracy. These results show that the body of politicians appointed by a dictatorship can contribute to the persistence of elites and institutions.
- ItemPannexin 1 regulates bidirectional hippocampal synaptic plasticity in adult mice(2014) Ardiles, Alvaro O.; Flores-Munoz, Carolina; Toro-Ayala, Gabriela; Cardenas, Ana M.; Palacios, Adrian G.; Munoz, Pablo; Fuenzalida, Marco; Saez, Juan C.; Martinez, Agustin D.The threshold for bidirectional modification of synaptic plasticity is known to be controlled by several factors, including the balance between protein phosphorylation and dephosphorylation, postsynaptic free Ca2+ concentration and NMDA receptor (NMDAR) composition of GluN2 subunits. Pannexin 1 (Panx1), a member of the integral membrane protein family, has been shown to form non-selective channels and to regulate the induction of synaptic plasticity as well as hippocampal-dependent learning. Although Panx1 channels have been suggested to play a role in excitatory long-term potentiation (LIP), it remains unknown whether these channels also modulate long-term depression (LTD) or the balance between both types of synaptic plasticity. To study how Panx1 contributes to excitatory synaptic efficacy, we examined the age-dependent effects of eliminating or blocking Panx1 channels on excitatory synaptic plasticity within the CA1 region of the mouse hippocampus. By using different protocols to induce bidirectional synaptic plasticity, Panx1 channel blockade or lack of Panx1 were found to enhance LIP whereas both conditions precluded the induction of LTD in adults, but not in young animals. These findings suggest that Panx1 channels restrain the sliding threshold for the induction of synaptic plasticity and underlying brain mechanisms of learning and memory.
- ItemThe Geography of Repression and Opposition to Autocracy(2023) Bautista, Maria Angelica; Gonzalez, Felipe; Martinez, Luis R.; Munoz, Pablo; Prem, MounuState repression is a prominent feature of nondemocracies, but its effectiveness in quieting dissent and fostering regime survival remains unclear. We exploit the location of military bases before the coup that brought Augusto Pinochet to power in Chile in 1973, which is uncorrelated to precoup electoral outcomes, and show that counties near these bases experienced more killings and forced disappearances at the hands of the government during the dictatorship. Our main result is that residents of counties close to military bases both registered to vote and voted "No" to Pinochet's continuation in power at higher rates in the crucial 1988 plebiscite that bolstered the democratic transition. Potential mechanisms include informational frictions on the intensity of repression in counties far from bases and shifts in preferences caused by increased proximity to the events. Election outcomes after democratization show no lasting change in political preferences.
